Gender-Bending
El Goonish Shive
Creator: Dan Shive
Format: Starts with strips, now pages.
Colors: Occasionally. Mostly grayscale.
Release Interval: Monday, Wednesday, Friday (with EGS:NP on Tuesday, Thursday, Saturday)

Set in Moperville, El Goonish Shive is probably the most unique comic I've ever seen. Not only does it have the most Gender-Bending of any comic I've ever read (It proves that, with a good enough storywriter, it is possible to gender-swap all the main characters at least once without it seeming like a crappy fanfic), it also has other types of transformation and abnormal stuff in abundance.
The main characters are Tedd, a nerd with a transformation gun (Gotta love those gender-based modes) and an effeminate face (He has 20/20 vision but wears big thick glasses to look more masculine), and Elliot (A fairly normal guy who knows martial arts and ends up getting transformed several times).
I don't want to reveal anything else for fear of spoiling it, but trust me... It's an amazing comic. If you've read enough Gender-bending manga to reach the point where it's starting to all feel the same (in a bad way), then give El Goonish Shive a try. If you haven't, give it a try anyway.
The release schedule is the standard Monday-Wednesday-Friday but there's also a subcomic called EGS:NP (El Goonish Shive: Newspaper) comprised of sidestories which may or may not make it into the main comic. EGS:NP is released Tuesday-Thursday-Saturday.
Finally, the flow of filler wavers around (1-7 pages per month) but, unlike some other comics (eg. like Megatokyo used to be), the filler is usually just as good as the main story (although not as relevant). Where else could you see such gems as all of the early El Goonish Shive characters' reactions to being bunnygirls or a sketch of "Darth Writers' Block"? Even the bland explanations are funny.

Ash wakes up one day, feeling like she would any other. There's just one problem, he went to bed a guy! Not only that, before she knows it, she has a new best friend who's missing the last two years of her life!
It turns out that it's all a celestial mix-up perpetrated by a pot-smoking angel and a little "Misfile". Thanks to the "Automatic Paradox Prevention", nobody else remembers the truth and if the other angels find out, things will get even worse.
The angel has assured them he can fix it if they give him some time, there's just one problem. Almost every attempt by Ash to protect his crumbling self-image is met by a new reason that her life may just be better.
It's the only webcomic I've seen so far with an "ask the main character questions" column. Just click Ask Ash in the header to see previous questions or ask a new one.
Venus Envy

Story about a pre-operative transgendered girl (male body, female self-image and mode of dress) trying to live her life in a fairly typical American environment. The characters are good and the illustration quality, though poor in the beginning, is perfectly good now.
I'd say more, but I don't want to spoil any surprises.
Venus Ascendant

The Venus Envy characters in a setting reminiscent of Star Trek. The story isn't half bad and the artwork, though it won't win any awards, won't hurt your eyes either.
Just Plain Funny

A humorous comic about a 20-year-old necromancer named Dave and Vyacheslav Molotov, who Dave resurrected. A Canadian creation and a funny read.
